Mejora de la termoestabilidad de la esterasa I de Pseudomonas fluorescens mediante selección de plegamiento in vivo en Thermus thermophilus

La estabilidad prolongada es una propiedad deseada para la aplicación biotecnológica de enzimas ya que permite su reutilización, contribuyendo a que los procesos biocatalíticos sean económicamente más competitivos con respecto a la síntesis química. En este estudio hemos aplicado selección por interferencia de plegamiento a alta temperatura en Thermus thermophilus para obtener variantes termoestables de la esterasa I de Pseudomonas fluorescens (PFEI). La variante más termoestable (Q11L/A191S) mostró una temperatura de fusión (Tm) de 77,3 ± 0,1 °C (4,6 °C más que el tipo salvaje) y una vida media de más de 13 horas a 65 °C (7,9 veces mejor que el tipo salvaje), con parámetros cinéticos sin cambios. Las mutaciones estabilizadoras Q11L y A191S se incorporaron a la variante L30P de PFEI, descrita previamente como enantioselectiva en la hidrólisis del enantiómero (-) de la lactama Vince. La variante final Q11L/L30P/A191S mostró una mejora significativa en la estabilidad térmica (Tm de 80,8 ± 0,1 °C y una vida media de 65 min a 75 °C), mientras conservaba la enantioselectividad (E > 100). Los estudios estructurales revelaron que A191S establece una red de enlaces de hidrógeno entre una horquilla en forma de V y el dominio de hidrolasa α/β que conduce a una mayor rigidez y, por lo tanto, contribuiría a explicar el aumento de la estabilidad.
